Age and Sex Differences in the Eμ-TCL1 Adoptive Transfer Mouse Model of CLL

چکیده

The Eμ-TCL1 model is the most widely used syngeneic mouse of chronic lymphocytic leukemia (CLL) and has been extensively to understand pathogenesis select genes, effect immune microenvironment for pre-clinical drug development studies. Recently there an increasing awareness impact age sex differences on cancers along with efficacy toxicity specific cancer therapies. However, despite predominance older males in CLL patient demographics, adoptive transfer (AT) studies have almost exclusively young female mice. We therefore sought how recipient survival order allow diverse using this model. First, a primary AT experiment was performed assess engraftment survival. found that AT, matched recipients had faster higher burden peripheral blood at 1- 3-months post-AT. while disease correlated survival, prolonged compared male mice regardless donor sex. Notably, were more prominent relatively indolent median 178 132 days respectively as aggressive donors where not apparent both 87 days. A subsequent secondary splenocytes isolated from showed course (median 56 days) which present. Additional experiments determine age. Three cohorts (young 3-6 months) middle aged (9-12 old (18-22 received AT. These ages selected correspond approximate human 20-30 years, 40-45 65 years old. While did significant rate establishment or mice, significantly decreased tumor engraftment, rates failure engraft, but similar overall younger cohorts. In conclusion, we show are present model, having particularly presentations. findings suggest must be considered experimental design adaptive CLL. p53-independent ibrutinib responses in an Eμ-TCL1 mouse model demonstrates efficacy in high-risk CLL

Deletion of the short-arm of chromosome 17 (17p-) is one of the most critical genetic alterations used in chronic lymphocytic leukemia (CLL) risk stratification. The tumor suppressor TP53 maps to this region, and its loss or mutation accelerates CLL progression, hampers response to chemotherapy and shortens survival. Although florescent in situ hybridization analyses for 17p deletions are routi...

Two types of BCR interactions are positively selected during leukemia development in the Eμ-TCL1 transgenic mouse model of CLL.

Chronic lymphocytic leukemia (CLL) is a common B-cell malignancy characterized by a highly variable course and outcome. The disease is believed to be driven by B-cell receptor (BCR) signals generated by external antigens and/or cell-autonomous BCR interactions, but direct in vivo evidence for this is still lacking. To further define the role of the BCR pathway in the development and progression...

miR-181b as a therapeutic agent for chronic lymphocytic leukemia in the Eμ-TCL1 mouse model

The involvement of microRNAs (miRNAs) in chronic lymphocytic leukemia (CLL) pathogenesis suggests the possibility of anti-CLL therapeutic approaches based on miRNAs. Here, we used the Eµ-TCL1 transgenic mouse model, which reproduces leukemia with a similar course and distinct immunophenotype as human B-CLL, to test miR-181b as a therapeutic agent. In vitro enforced expression of miR-181b mimics ...

The Syk inhibitor fostamatinib disodium (R788) inhibits tumor growth in the Eμ- TCL1 transgenic mouse model of CLL by blocking antigen-dependent B-cell receptor signaling.

Inhibition of antigen-dependent B-cell receptor (BCR) signaling is considered a promising therapeutic approach in chronic lymphocytic leukemia (CLL), but experimental in vivo evidence to support this view is still lacking. We have now investigated whether inhibition of BCR signaling with the selective Syk inhibitor fostamatinib disodium (R788) will affect the growth of the leukemias that develo...
